Galera Therapeutics entered 2026 focused on an acquired nitric-oxide-synthase inhibitor for difficult-to-treat breast cancers while pursuing a strategic combination with Obsidian Therapeutics. Tilarginine Pan-NOS Program
Tilarginine is a pan-inhibitor of nitric oxide synthase. The program is based on evidence that nitric oxide signaling can support tumor initiation, progression, metastasis, and immune suppression in certain cancers.
Metaplastic and Triple-Negative Breast Cancer
The lead Galera development focus is metaplastic breast cancer and other refractory subsets of triple-negative breast cancer, where nitric oxide synthase can be overexpressed and treatment options are limited.
Investigator-Sponsored Combination Study
An NIH-funded Phase 1/2 investigator-sponsored trial is evaluating tilarginine with nab-paclitaxel and alpelisib in metaplastic breast cancer. A separate triple-negative breast cancer study has also been contemplated through the I-SPY 2 consortium, subject to funding and development decisions.
Divested Dismutase-Mimetic Assets
Galera sold avasopasem, rucosopasem, and its other superoxide-dismutase-mimetic assets to Biossil. This transaction removed the company's former radiotherapy-support portfolio and left tilarginine as its principal legacy development asset.
Obsidian Combination and OBX-115
The strategic combination with Obsidian centers the combined company on OBX-115, an engineered autologous tumor-infiltrating lymphocyte therapy designed to produce regulated membrane-bound IL-15. OBX-115 is in clinical development for advanced melanoma and non-small cell lung cancer.
