Genetically Defined Diseases
Cogent Biosciences develops precision therapies for diseases driven by defined oncogenic mutations. Its portfolio is led by a selective KIT inhibitor and includes earlier programs directed at ErbB2, PI3Kα, KRAS and JAK2.
Bezuclastinib
Bezuclastinib is designed to inhibit KIT D816V and other exon 17 KIT mutations while limiting off-target activity. These mutations drive systemic mastocytosis and are also important resistance mechanisms in gastrointestinal stromal tumors.
Systemic Mastocytosis Franchise
The company is pursuing bezuclastinib in both non-advanced and advanced systemic mastocytosis. An NDA for non-advanced disease is under FDA review, while the APEX program supports a separate planned application in advanced disease.
Gastrointestinal Stromal Tumors
The Phase 3 PEAK trial supports an NDA for bezuclastinib in GIST after prior imatinib treatment. Cogent also plans to study bezuclastinib with sunitinib in first-line exon 9-mutant GIST, potentially extending use into an earlier treatment setting.
